Bacterial Periplasmic Binding Proteins

چکیده

The cell envelope of gram-negative bacteria includes inner and outer membranes with the periplasmic space in between. The transport systems for many nutrients in these organisms include components in both the periplasm and the inner membrane. In some cases, e.g., maltose, transport components in the outer membrane are also involved. The scope of this review will be limited to a progress report on our understanding of the nature and the role of the periplasmic components that serve as receptors or binding proteins (BPs) * for the various nutrients. Bacterial periplasmic BPs for a large variety of carbohydrates, amino acids, inorganic ions, and enzyme cofactors have been isolated and characterized. More complete reviews on these BPs have appeared recently (Oxender and Quay, 1976; Wilson and Smith, 1978; Furlong and Schellenberg, 1980).

IMPROVEMENT OF hGM-CSF EXPRESSION USING A FUSION SYSTEM BY DIRECTING PROTEIN TO THE PERIPLASMIC SPACE

Bacterial expression systems provide economic and logistic advantages in manufacturing proteins for human therapeutic purposes. However, most such proteins accumulate in insoluble biologicaIIy inactive form when overexpressed in bacterial cells. This is the case while attempting to produce recombinant human granulocyte-macrophage colony stimulating factor (hGM-CSF) in E. coli. In this repor...

Characterization of the ligand-binding domains of glutamate receptor (GluR)-B and GluR-D subunits expressed in Escherichia coli as periplasmic proteins.

We recently reported that a functional ligand-binding site of an alpha-amino-5-methyl-3-hydroxy-4-isoxazole propionate (AMPA)-selective glutamate receptor (GluR)-D subunit can be expressed in insect cells as a soluble, N-glycosylated fusion protein consisting of two segments (S1 and S2) that are related by amino acid sequence to bacterial periplasmic binding proteins (Kuusinen, A., Arvola, M., ...

Identification of the Amino Acid Subsets Accounting for the Ligand Binding Specificity of a Glutamate Receptor

In a situation so far unique among neurotransmitter receptors, glutamate receptors share amino acid sequence similarities with the bacterial periplasmic binding proteins (PBPs). On the basis of the primary structure similarity of two bacterial periplasmic proteins (lysine/arginine/ornithine- and phosphate-binding proteins) with the chick cerebellar kainate-binding protein (KBP), a member of the...

Structure-based design of a periplasmic binding protein antagonist that prevents domain closure.

Many receptors undergo ligand-induced conformational changes to initiate signal transduction. Periplasmic binding proteins (PBPs) are bacterial receptors that exhibit dramatic conformational changes upon ligand binding. These proteins mediate a wide variety of fundamental processes including transport, chemotaxis, and quorum sensing. Despite the importance of these receptors, no PBP antagonists...

Distinguishing multiple chemotaxis Y protein conformations with laser-polarized

The chemical shift of the Xe NMR signal has been shown to be extremely sensitive to the local environment around the atom and has been used to follow processes such as ligand binding by bacterial periplasmic binding proteins. Here we show that the Xe shift can sense more subtle changes: magnesium binding, BeF3 − activation, and peptide binding by the Escherichia coli chemotaxis Y protein. H-N c...
